1.病毒感染和宿主免疫及肿瘤发生中的蛋白质脱酰胺化修饰研究

蛋白质脱酰胺化修饰在微生物感染与致病过程中的作用已经逐渐被人们所认识。我们在前期工作的基础上,开展对禽流感、中东呼吸综合症、寨卡和疱疹病毒感染和宿主免疫过程中的蛋白质脱酰胺化修饰的调控作用的研究,探究病毒劫持宿主的代谢酶进行蛋白质脱酰胺化修饰的调控机制,以及这种调控对病毒感染和宿主免疫的生理和病理意义。同时我们也将探究这种修饰在恶性肿瘤发生中的调控作用。该项工作与美国南加州大学的冯平辉教授进行深度合作。

2.肿瘤发生中泛素化蛋白的蛋白质组学研究

探索并运用新的蛋白质组学方法对肿瘤组织、肿瘤细胞和肿瘤干细胞的泛素化蛋白进行全蛋白质组学(global proteomics)分析,发现新的泛素化修饰底物及潜在的分子标记。

3.肿瘤免疫治疗之CAR-T和TCR-T细胞治疗技术的开发和应用

CAR-T技术是目前最有前途的肿瘤免疫治疗新策略,对血液肿瘤的治疗特别有效,然而对实体瘤的效果有限。研究组将对现有的技术进行改造,以期在实体瘤的CAR-T治疗方向取得突破。另外由于TCR-T技术在实体瘤治疗的临床试验中显示了很好的疗效,实验室通过与企业合作,联合开发TCR-T细胞治疗技术。
